چکیده

BACKGROUND AND PURPOSE Predicting the risk of drug-induced adverse psychiatric effects is important but currently not possible in non-human species. We investigated whether the affective bias test (ABT) could provide a preclinical method with translational and predictive validity. EXPERIMENTAL APPROACH The ABT is a bowl-digging task, which quantifies biases associated with learning and memory. Rats encounter independent learning experiences, on separate days, under either acute manipulations (e.g. pro-depressant vs. control) or different absolute reward values (e.g. high vs. low). A bias is observed during a preference test when an animal's choices reflect their prior experience. We investigated the effects of putative pro-depressant drug treatments following acute or chronic administration on the formation of an affective bias or reward-induced positive bias respectively. KEY RESULTS The immunomodulators LPS (10 μg·kg-1 ), corticosterone (10 and 30 mg·kg-1 ) and IFN-α (100 U·kg-1 ) induced a negative affective bias following acute treatment. Tetrabenazine (1 mg·kg-1 ) also induced a negative bias, but no effects were observed with varenicline, carbamazepine or montelukast. Chronic treatment with IFN-α (100 U·kg-1 ) and retinoic acid (10 mg·kg-1 ) impaired the formation of a reward-induced positive bias but did not alter sucrose preference test (SPT). CONCLUSIONS AND IMPLICATIONS The ABT has the potential to provide a novel approach to predict pro-depressant risk in a non-human species. Negative biases induced by acute treatment in the standard version of the task may also predict longer-term effects on reward processing as shown by the deficit in reward-induced positive bias following chronic treatment, an effect distinct from anhedonia in the SPT. تأثیر آموزش تنظیم عاطفه بر عواطف مثبت و منفی زنان مبتلاء به اختلال ناشی ازمصرف متامفتامین

Background & Aims: Although affective disorders and substance use disorders (SUDs) frequently co-occur, the role of affective dysregulation in addiction is often overlooked. Individuals with affective psychopathology have been shown to demonstrate high rates of comorbid SUDs. These findings have suggested that the PA and NA systems may differentially mediate different aspects of substance ...

Exploring Neural Correlates of Different Dimensions in Drug Craving Self-Reports among Heroin Dependents

Introduction: Drug craving could be described as a motivational state which drives drug dependents towards drug seeking and use. Different types of self-reports such as craving feeling, desire and intention, wanting and need, imagery of use, and negative affect have been attributed to this motivational state. By using subjective self-reports for different correlates of drug craving along ...

Human Laboratory Settings for Assessing Drug Craving Implications for the Evaluation of Treatment Efficacy

Research on assessing craving in laboratory settings often involves inducing and then measuring craving in subjects. Cue-induced craving is studied in laboratory settings using the cue reactivity paradigm, in which drug-related photos, videos, evocative scripts, olfactory cues, and paraphernalia may induce craving. Cue-induced craving evoked by drug-related stimuli could be associated with rela...

The plasticity of adolescent cognitions: data from a novel cognitive bias modification training task.

Many adult anxiety problems emerge in adolescence. Investigating how adolescent anxiety arises and abates is critical for understanding and preventing adult psychiatric problems. Drawing threat interpretations from ambiguous material is linked to adolescent anxiety but little research has clarified the causal nature of this relationship. Work in adults using Cognitive Bias Modification of Inter...
